Professor Layton attends an opera performance at the invitation of the celebrated singer Jenis Quatlane, who wants his help with a strange mystery. A year ago, her friend Melina died. Now a seven-year-old girl has turned up saying she has discovered the secret of eternal life, and she knows things only the real Melina could know. Layton and his apprentice Luke travel to the Crown Petone opera house, while Remi stays behind in London to look into the disappearance of several girls. The opera tells of a lost kingdom whose people turned immortal after their beloved queen died, hoping to see her return. When the curtain falls, a masked figure announces a deadly game: the winner gains eternal life, and the losers die. Layton and Luke must play along and uncover the truth behind the promise of immortality in this mystery film spun off from the puzzle games.
