Lupin III, Goemon and Jigen fly a mini-helicopter to the mysterious Drifting Island, chasing rumors of a treasure hidden somewhere on it. While exploring, the trio sets off the alarm and wakes the Nanomachine, the island's deadly security system. The key to the island's mystery lies in Zufu, a small nation that was once prosperous and is now ruled by General Headhunter, a ruthless man who collects knives. Fujiko hacks into the general's computer in search of vital information. Meanwhile, Zenigata receives a video message in which Lupin declares he is after the treasure, and Oleander, a fiery blond officer with secrets of her own, joins him. With what they have learned, the thieves head back to the island, this time with General Headhunter on their trail. The result is a comedic adventure caper.
